授業概要
Course overview
The objective of this course is for students to improve the understanding of the anatomical features
and functions of the respiratory system from the point of view of integrated physiology. Students will
learn several techniques related to molecular biology and genetic epidemiology to elucidate etiologies and pathophysiological
processes of respiratory diseases and to develop novel therapeutic options for them. Furthermore, this
course provides a brief review of skills related to a clinical study for designing and conducting translational
research.        
The goal of this seminar is for students to improve the critical skills of reading scientific papers to understand the objectives, methods and findings of the research, which are then discussed by the group members to elucidate its significance and limitations. Students will also discuss how to design and conduct experiments to obtain skills that are needed to draw up their own research plans. The goal of this course is for students to learn practically about research principles and methods including life-science techniques to elucidate the etiology and pathophysiology of respiratory diseases. Students will also learn how to analyze and interpret the results of the experiments. Furthermore, through this course, we aim to provide students with research skills and experience that prepare them to be capable of pursuing and developing their subsequent studies on their own.

Upon successful completion of this course, the student will be able to:
1. Understand the anatomy and physiology of the respiratory system, and explain the etiologies and
pathophysiological processes of a wide range of respiratory diseases.
2. Explain appropriate diagnostic and therapeutic options for a wide range of respiratory diseases.
3. Understand various experimental approaches such as molecular biological and genetic
techniques, and apply them to the research projects in the field of pulmonology.
4. Understand ethical and statistical aspects of clinical studies, and plan and carry out researches
properly.
Upon successful completion of this seminar, the student will be able to:
1. Search, collect and select appropriate scientific papers presenting novel findings on etiology, pathogenesis, diagnostic techniques, or treatment options of respiratory diseases.
2. Explain the scientific background of the research.
3. Explain the scientific characteristics and originality of the research in the field.
4. Understand and explain the methods used in the research and interpretations of the presented findings.
5. Explain the scientific significance and limitations of the research.
1. Obtain clinical information and specimens such as blood, bronchoalveolar lavage fluid and tissue for molecular biological and genetic research.
2. Extract genomic DNA or RNA from whole blood, tissue and cultured cells.
3. Detect and analyze genetic polymorphism (e.g., allele-specific PCR, DNA sequencing, restriction fragment length polymorphism, etc.).
4. Perform statistical analysis on the association of genetic polymorphism with disease phenotypes.
5. Breed wild-type and genetically modified mice, establish appropriate animal models, and obtain biological specimens for use in research.
6. Passage, freeze and thaw cultured cells.
7. Prepare formalin-fixed paraffin-embedded samples, cut them in thin sections, perform appropriate staining, and observe the stained sections under light microscopes.
8. Detect and analyze the expression of proteins by western blotting.
9. Detect and analyze the expression of mRNA by real time PCR and RNA-seq.

Grading Criteria
Students are evaluated by the achievement of the objectives listed in the SBO.
- Good achieved under supervision in the SBOs 1 and 2:  Pass (C).
- Good achieved under supervision in the SBOs 1 - 3, and actively conducted SBO 4:  Pass (B).
SBOs 1 - 3 will be evaluated based on a brief report in terms of presentation and dicsussion.
- Excellent achievement in SBOs 1-4: Pass (A)
- Outstanding achievement with enough evidence in all SBOs : Pass (A+)
Students are evaluated by the achievement of the objectives listed in the SBO.
- Good achieved  in the SBOs 1 and 2:  Pass (C).
- Good achieved in the SBOs 1 - 2, and actively conducted SBO 3:  Pass (B).
Based on written summary of papers (A4 1 page, twice a month).  good achievement including SBO 4 is confirmed,: Pass (A).
- Outstanding achievement with enough evidence in all SBOs : Pass (A+)
Skills to obtain reliable experimental results (50%)
Understanding of results, and ability to design next experiments will be evaluated by experimental notebook and discussion (50%)
A+: Superior (more than 90: top 10%)
A: Excellent (80-89: upper 20%)
B: Good (70-79)
C: Average (60-69)
D: Failure (less than 60)
