Synaptic Strength in Prefrontal Cortex Regulates Homeostatic Sleep Need

要旨
Although sleep is crucial for memory consolidation, the link with synaptic strength remains undetermined. We explored a causal relationship between cellular synaptic strength and macro-level sleep pressure, as indicated by EEG delta power. Our mathematical model introduced a mechanism: increased synaptic strength promotes neuronal hyperpolarization and raised delta power, which was validated using primary cultured neurons in vitro. Moreover, our novel molecular tool (SYNCit-K), which induces dendritic spine enlargement and synaptic potentiation via chemically induced dimerization, demonstrated that synaptic potentiation of the prefrontal cortex's (PFC's) excitatory neurons but not the primary visual cortex or inhibitory neurons, increased EEG delta power and NREM sleep amounts. In summary, PFC excitatory neurons' synaptic strength dictates sleep pressure in mammals.
