Elucidating the Pathogenesis of Heritable Aortic Diseases: Insights from Marfan Syndrome
要旨 Abstract:
Heritable aortic diseases, including Marfan syndrome, can lead to life-threatening thoracic aortic aneurysms and
dissections. Despite advances in human genetics that have led to the identification of causative genes, the
mechanisms underlying disease onset and progression remain poorly understood, limiting the development of novel
pharmacological therapies. We recently identified a novel point mutation in FBN1, which encodes the large
extracellular matrix protein fibrillin-1, in a patient with aortic dissection. By introducing this point mutation,
we established a novel mouse model that develops severe, early-onset aortic dissection. By integrating single-cell
transcriptomic analysis with this model, we have identified dynamic inflammatory responses that may contribute to
disease pathogenesis. In this presentation, I will provide an overview of heritable aortic diseases and highlight
some of our recent findings.
