Analysis for transcriptional regulation on early stage of
iPS cell production

During iPS cell production, first, somatic cells-specific gene expression is decreased, and then, genes for pluripotency are expressed. Because of such a broad gene expression change, transcription factors are considered to have important roles to iPS cell production. However, as a result of low efficiency of conventional iPS cell production methods, many insufficient reprogrammed cells with no/low pluripotency are contaminated in iPS cells. It makes difficult to investigate the mechanism of iPS cell production especially in early stage. Our iPS cell production methods are very efficient compared to conventional one, so they are used for identifying transcription factors relating to early step of cell reprogramming. And we investigate molecular mechanism of cell reprogramming by focusing on functions of these transcription factors. These analyses will make us enable to establish methods of more efficient production of safe iPS cells.
